Description
Lack of ability to develop vascularized thick tissue constructs is a major limitation for use in tissue repair and regeneration in humans. Even though tremendous advancements have been developed over the last decade, significant gaps remain in replicating the in vivo scenario. A key bottleneck, of particular relevance to thick tissues is the mass transfer limitations across the vasculature to the tissue cells, which detrimentally impacts the long term viability and functionality of the tissue constructs. In this context, we propose to develop (design, fabricate, test, and demonstrate) a novel platform for generation of vascularized thick tissue constructs that mimick the desired cell, tissue and organ architecture. During Phase I, we will demonstrate proof-of-concept using a synergistic in silico and in vitro approach to guide the development of the thick vascularized tissues. Both viability and biomarker analysis will be performed to demonstrate viable tissues. In Phase II, we will further characterize the tissue constructs by functional assays and long term maintenance. The developed constructs will be validated against in vivo data for both structural and functional responses. Finally, we will expand the system to mimic additional organs of interest. Process of FDA clearance will be initiated.
