Project Summary The overall goal of this program is to develop a nanopore that will allow DNA mononucleotides such as those released from an exonuclease to be captured and read with accuracy thus enabling the exonuclease nanopore based sequencing concept While base discrimination with a molecular adapter has been previously demonstrated capturing the molecules with efficiency is the current limitation By creating the proposed device we will be able to use novel methods to control the molecular transport rate of the mononucleotides to and through the pore enabling capture efficiency Project Narrative The proposed program will address critical factors currently preventing nanopore sequencing from realizing its full potential The success of this program will thus help enable rapid and inexpensive DNA sequencing which will greatly increase access to full genome sequencing Such routine DNA sequencing will enable significant improvements for human health including the use for personalized medicine detecting and understanding genetic mutations and diseases as well as the use with pathogen detection
