Description
Rapid sensitive and specific diagnosis of the Dengue virus DENV infection is important for both patient care and epidemiologic studies Current Dengue diagnostic tests are either time consuming expensive or lack of sensitivity We propose to develop a novel low cost lateralflow immunoassay LFIA strip capable of sensitive detection of DENV NS antigen The proposed test strip synergically integrates the simplicity of LFIA and the high sensitivity of Surface enhanced Raman scattering SERS detection by incorporating novel dye embedded Au@silica core shell nanoparticles In addition this novel LFIA is a dual mode detection strip which the assay results can be detected by using portable Raman or fluorescent reader In phase Iproject we will synthesis and characterization of the dye embedded Au@silica nanoparticles as extinct Raman label design and fabricate LFIA strip incorporates the nanoparticles demonstrate the strip for sensitive detection of NS determine the sensitivity and limit of detection in simulated clinical samples and validate the strip with current commercial NS test In Phase II project we will develop LFIA strips capable of detection of multiple DENV biomarkers of NS lgM and lgG in the samples of serum plasma and blood and validate the strip with large volume of clinical samples
